Abstract:
This data set describes nuclear microsatellite genotypes derived from eight autosomal loci (BCA6, BCA9, BCA11, BCA5, CRG, OXY13, TSP1.20.09 and TSP1.20.46) and a 366 base pair portion of the mitochondrial control region. A total of 665 Greater White-fronted Geese were examined by genotyping and 518 by sequencing with samples coming from its circumpolar range representing multiple flyways (Midcontinent, Pacific, Greenland, Palearctic). The sex of most samples was determined in the field by morphology or cloacal examination.
Purpose:
To examine nuclear microsatellite allele variation and population genetic structure among samples of Greater White-fronted Geese (Anser albifrons) across its circumpolar range.

Individual sample ID's, collection dates, sampling locations, band numbers, and reported sex of individual samples provided by collectors were cross-checked and verified to match the accompanying worksheet. Sampling locations were verified with collectors and formally labelled and cross-referenced to the Geographic Names Information Service (GNIS) or NGA GEOnet Names Server when possible. Species identification by collectors were informally verified by confirming genetic "signatures" (e.g. microsatellite size or DNA sequence) fell within expected values for the species. Field sex was not confirmed with a laboratory analysis. Genbank accession numbers were cross-referenced to the appropriate markers in attribute definitions. For quality control purposes, all genotypes were scored relative to known size standards run on each gel. These size standards were established by running them with an M13 DNA sequence ladder of known size. Additionally, ten percent of the samples were amplified and genotyped in duplicate for the 8 microsatellite loci to double-check genotype scores. Final genotype scores were reviewed by a second laboratory specialist.

For microsatellite data, reported allele sizes were reviewed to confirm they conform to expected distributions (e.g. dinucleotide repeats with 1 base pair differences were reviewed on gel images with laboratory personnel, gaps in allele sizes were reviewed, and single occurrences of an allele size were scrutinized).

Data omissions for particular entities are explained in the Entity and Attribute definitions. Blank data indicates no data were collected or not provided with the sample for a particular attribute. Attribute data equaling “NP” in a cell of the data set refers to a sample that did not amplify or show any product (“no product”). Not all samples originated from a banded bird. If known, the band number has been provided.

No formal positional accuracy tests were conducted. The coordinates of samples are for general purposes only. The coordinates only designate a general regional location for each population and should not be considered at exact location of each sample. Coordinates were determined in Google Earth by searching general location information. For example, Hook Lagoon, Alaska Peninsula was searched and the coordinates from google earth was assigned to those samples.

Polymerase chain reaction (PCR) amplification of mtDNA followed Sonsthagen et al (2004) using primer pairs as published [WFGL1M (Ely et al., 2017), H542 (Sorenson and Fleischer, 1996). PCR amplifications were carried out in a 20-μL volume with 2–100 ng genomic DNA, 0.5 μM each primer, 1.0 μM dNTPs, 10× PCR Perkin Elmer Cetus #1 buffer, and 0.2 units Taq polymerase. PCR reactions began with 94 ºC for 10 min followed by 40 cycles of 94°C for 30 sec, 50 °C for 30 sec, and 72 °C for 90 sec with a 30 min final extension at 72 °C. ExoSAP-IT® (USB Corporation, Cleveland, OH) was used to remove excess primers and dNTPs in PCR products. All samples were cycle-sequenced on a LI-COR 4200LR yielding a 366 base pair fragment for all samples.

Genotype numbers for all loci were determined by scoring based on PCR product size relative to an M13 DNA sequence ladder using Gene Profiler 4.05 (Scanalytics, Inc.). Subsequent gels included these known size individuals from which relative sizes of unknown individuals were assigned using Gene Profiler 4.05 (Scanalytics, Inc.). MtDNA control region sequences from opposite strands were reconciled using LI-COR eSeq imaging software with visual proofing and aligned using AlignIR 2.0 (LI-COR, Inc., Lincoln, NE) or Sequencher 4.1.2 (Gene Codes Corporation, Ann Arbor, MI).
