This data set contains genetic information collected from eelgrass (Zostera marina) populations along the Pacific coast of North America from Alaska to Baha California. A total of 447 samples were collected comprising 401 unique individuals (genets) and 46 clones (ramets) from which 10 microsatellite DNA loci were obtained.
- Purpose:
-
The central aim of this study is to characterize genetic structure in largely undisturbed natural populations of eelgrass along the northwestern-most Pacific coast of North America. Data from microsatellite DNA loci were used to (1) quantify levels of genetic diversity and structuring within and among natural undisturbed populations of Z. marina along the North Pacific and Bering Sea coasts of Alaska, which are geographically separated by the Alaska Peninsula; (2) test for polarity of gene flow among populations within the Eastern Bering Sea Large Marine Ecosystem and the Gulf of Alaska Large Marine Ecosystem; and (3) test for association between genetic and latitudinal gradients among populations along the Pacific coast of North America.

Genetic markers used are as follows: Data include fragment data from 1) 10 microsatellite DNA loci from 447 samples comprising 401 unique individuals (genets) and 46 clones (ramets) sampled from populations along the Pacific coast of North America, including Oregon (Yaquina Bay, n = 25 genets and 6 ramets) and Alaska: Saint Catherine’s Cove (n = 24 genets), Kinzarof Lagoon (n = 48 genets), Izembek Lagoon (n = 71 genets), Wide Bay (n = 33 genets), Safety Lagoon (aka Safety Sound; n = 34 genets), Unga Island (n = 6 genets and 4 ramets), Simeonof Island (n = 6 genets and 1 ramet), Nakwasina Bay (n = 30 genets), Kuskokwim Shoals in the Kuskokwim Bay (n = 29 genets and 12 ramets), Montague Island, Prince William Sound (n = 46 genets, 22 ramets), Togiak Bay (n = 49 genets, 1 ramet); and 2) 6 loci from 138 samples (genets only) collected along a latitudinal gradient from the southern portion of the species’ range (Baja California: Estero Punto Banda, n = 37; Bahia San Quintin, n = 40, Bahia Magdalena, n = 28); and the middle Pacific (California: Monterey Bay, n = 10; Washington: Shallow Bay in Puget Sound (n = 23).

Polymerase chain reaction (PCR) amplification and electrophoresis followed protocols described in Sonsthagen et al. (2004; Sonsthagen, S.A., Talbot, S.L., and White, C.M. 2004. Gene flow and genetic characterization of Northern Goshawks breeding in Utah. Condor 106:826–836.).
Genotype numbers were determined by scoring based on allele size relative to an M13 DNA sequence ladder and to samples established as size standards that were run on each gel.
For quality control purposes, 10% of the samples were extracted, amplified, and genotyped in duplicate. We observed sterile technique in the handling of all DNA, and all procedures were performed with positive and negative controls to provide evidence of replication without contamination.
