Abstract:
This data set contains allele sizes for 30 individuals of 9 previously published and 20 novel microsatellite loci and the molecular sex verification (n = 31) of Emperor Geese (Chen canagica) from various locations across Alaska. The allele sizes for the 20 microsatellite loci developed in this study are included for 10 White-fronted Geese (Anser albifrons) from Selawik, 11 Black Brant (Branta bernicla nigricans) from Kigigak Island, 10 Canada Geese (Branta canadensis) from Copper River Delta, and 9 Cackling Geese (Branta hutchinsii) from lower Kashunuk River (Hock Slough) and Tutakoke River.
Purpose:
Previously published microsatellites (n=9), novel microsatellites developed by the authors (n=20 including 1 juxtaposed), and a diagnostic sexing marker were screened and characterized for the usefulness of individual ID and population characterization of the Emperor Goose (Chen canagica). The twenty novel microsatellites were also screened for variability in 4 other goose species.

Individual sample ID's, collection dates, sampling locations, band numbers, and reported sex of individual samples provided by collectors were cross-checked and verified to match the accompanying worksheet.
Sampling locations were verified with collectors and formally labelled and cross-referenced to the Geographic Names Information Service (GNIS) when possible.
All HINT sex identifications matched anatomical sex recorded at time of necropsy.
Species identification by collectors were informally verified by confirming genetic "signatures" (e.g. microsatellite size or DNA sequence) fell within expected values for that species.
Genbank accession numbers were cross-referenced to the appropriate individual samples as recorded.
For quality control purposes, DNA from 18-20% of individuals (n = 11-18 Emperor Geese and n = 2 for each of the other species) were extracted and microsatellites amplified in duplicate. DNAs from 7 Emperor Geese (22%) were extracted and amplified for the HINT gametolog marker in duplicate. All duplicate characterizations of the same individual resulted in concordant data.
If applicable, all but one of multiple samples characterized by identical composite genotypes across all loci were eliminated.

For microsatellite loci, reported allele sizes were reviewed to confirm that they conform to expected distributions (e.g. dinucleotide repeats with 1 base pair differences were reviewed on gel images by laboratory personnel, gaps in allele sizes were reviewed, and single occurrences of an allele size were scrutinized).
Completeness_Report:
Data omissions for particular entities are explained in the Entity and Attribute definitions. Blank data indicates no data were collected or not provided with the sample for a particular attribute. Attribute data equaling "NP" in a cell of the data set refers to a sample that did not amplify or show any product ("no product"). Not all samples originated from a banded bird. If known, the band number has been provided. Field sex was not confirmed with a laboratory analysis for species other than Emperor Goose for this study and has not been provided. HINT_Z Genbank accession numbers are present only for the samples used for HINT_Z marker development of a fragment based system. Emperor Geese with no HINT_Z accession numbers were sexed via the fragment based system only. Microsatellite loci developed in other waterfowl species (Aph2, Bca4, Bca5, Bca11, Oxy13, Smo01, Smo10, Smo11, and Smo13) and used in prior analyses of Emperor Geese (Watkins V.S. 2006. Intraspecific nest parasitism in Emperor Geese (Chen canagica): a population genetic analysis. M.Sc. Thesis, Alaska Pacific University) were not evaluated in other species of geese for this study. Five Emperor Geese with no microsatellite data were used only to develop the Hint_Z sexing marker.
