Description:
Abstract:
This dataset consists of one table with diagnostic screening results from a study of Francisella tularensis bacteria exposure in tundra-nesting geese and mammals and Brucella spp. exposure in mammals sampled from multiple locations on the Arctic Coastal Plain of Alaska, 2014-2017. The table provides sample collection and location data of geese, arctic fox, red fox, caribou, small mammal, and polar bear samples.
Purpose:
These data provide the basis to: 1) investigate the seroprevalence of F. tularensis antibodies in wildlife sampled on and near the Arctic Coastal Plain of Alaska, 2) to investigate the general patterns of F. tularensis and Brucella spp. exposure in this region.

ITIS currently lists Snow Geese in the genus Chen. However, the most current taxonomic designation by the American Ornithological Society's (AOU) Check-list of North American Birds lists Snow Geese in the genus Anser. Here, we follow the current AOU designation as described in: Chesser, R.T., Burns, K.J., Cicero, C., Dunn, J.L., Kratter, A.W., Lovette, I.J., Rasmussen, P.C., Remsen, J.V., Jr., Rising, J.D., Stotz, D.F., Winker, K. 2017. Fifty-eighth supplement to the American Ornithological Society's Check-list of North American Birds. The Auk:134(3)751-773. doi:10.1642/AUK-17-72.1
Taxonomic_Procedures:
Species were identified by skilled observers in the field based on general appearance, plumage, pelage, behavior, and vocalization.
Taxonomic_Completeness:
Taxonomy is complete for all samples. No vouchers were collected.

Attribute_Accuracy_Report:
Collection sites, sampling data, and field and lab taxa were all cross-checked and verified to match field data. Individual laboratory ID numbers, species, age, and sex data were verified to be correct according to field ID numbers. Sterile technique was used during serological screening, including the use of positive controls and blind replicate testing. All reported serologic data and corresponding titers were cross-checked and confirmed to be correct.
Logical_Consistency_Report:
Attribute values fall within expected ranges and data were proofed for the presence of duplication and omission.
Completeness_Report:
No data were omitted and there are no missing data. Blank cells indicate that no serological screening test for F. tularensis or Brucella bacteria was performed on that respective sample or that no GPS location data was recorded for that respective sample.

Methodology:
Methodology_Type: Field
Methodology_Description:
Blood samples were collected by USGS researchers or partners during field capture efforts and serum was isolated on the same day of collection. Sera was frozen in the field and transported on ice to the USGS Alaska Science Center where it was stored at -80 C until subsequent laboratory analysis. Additional details regarding field sampling are available in Smith et al. 2022.
Methodology:
Methodology_Type: Lab
Methodology_Description:
Sera was tested for the presence of F. tularensis antibodies using a commercially available febrile antigen slide agglutination test (FAAT; Becton, Dickinson and Company, Sparks, MS, USA). Serial dilutions of sera ranging from 1:20-1:320 were tested in parallel with F. tularensis antisera to serve as positive controls. Titers were scored based on the amount of agglutination present in each dilution during a visual scan 60 seconds after mixing. All titers that produced visible agglutination scored at ≥1:20 compared to positive controls were interpreted as containing F. tularensis antibodies. Additional details regarding lab processing are available in Smith et al. 2022.
Methodology:
Methodology_Type: Lab
Methodology_Description:
Samples were initially screened for Brucella abortus using buffered, acidified plate antigen (BAPA) followed by confirmation with fluorescence polarization assay (FPA). Only samples positive by both BAPA and FPA (≥20 mP) were considered positive. A 2-Mercaptoethanol Tube Agglutination Test (MAT) was used to screen samples for B. canis (titer greater that 50 is considered positive, titer = 50 considered suspect). Additional details regarding lab processing are available in Smith et al. 2022.
