Abstract:
This dataset is three tables with details of samples and aliquots of waterfowl feces deposited in filtered surface water collected from an urban waterbody in Anchorage, Alaska in 2018-2019. Sample vials were submerged underwater in the same waterbody from which the samples were collected and the samples were tested for the presence and viability of influenza A virus. Temperature data and water chemistry measurements were also collected over time.

Water was collected at Cuddy Family Midtown Park pond in Anchorage, Alaska and passed through a series of 1 micron and 0.22 micron filters.

Environmental Fecal Sampling: Swab samples for viral screening were collected by applying a polyester-tipped applicator to the feces. All swabs were placed in cryovials with chilled filtered surface water from Cuddy Family Midtown Park pond in Anchoage Alaska. Twenty aliquots were made from each sample and all but one aliquot from each sample were placed in mesh bags weighted with rocks and sealed in steel drums in approximately one meter of water.

Environmental Fecal Sampling: All swab samples were initially screened at the University of Georgia using real-time reverse transcriptase polymerase chain reaction (rRT-PCR) targeting gene products for the influenza A virus matrix gene. All samples yielding Ct values < 45 for the IAV matrix gene were inoculated into specific pathogen-free embryonated chicken eggs for virus isolation. Amnio-allantoic fluids were collected after 120 hours and tested via hemagglutination assay. RNAs extracted from hemagglutination assay positive amnio-allantoic fluids were tested for the IAV matrix gene by rRT-PCR and resultant isolates were genomically sequenced at the Alaska Science Center using next-generation sequencing technology (Illumina platform, Illumina Inc., San Diego, CA).
