Abstract:
This data set contains information on blood chemistry and hematological parameters of captive Pacific Common Eiders (Somateria mollissima v-nigrum) for reference periods and after satellite transmitter implantation.
Purpose:
These data were collected for health assessments, to evaluate seasonal changes in regularly measured biomarkers, to assess the physiological response to dive conditioning, and to measure responses to surgical implantation of a satellite transmitter for captive Pacific Common Eiders.
Supplemental_Information:
This data set contains seven tables of blood chemistry and hematological information assessed using blood samples from captive Pacific Common Eiders. These data were collected for health assessments, to evaluate seasonal changes in regularly measured biomarkers, to assess the physiological responses to dive conditioning, and to measure responses to surgical implantation of a satellite transmitter. All data presented for creatine kinase and data from 09/29/2005 to 03/15/2006 for aspartate aminotransferase, glucose, lactate dehydrogenase, and manual packed cell volume are summarized in Latty et al. (2016) describing physiological responses to transmitter implantation. Serum protein and white blood cell proportions and serum total protein using the biuret method are also utilized in Latty et al. (2016), but not directly reported [i.e., ratios are reported for hetrophils to lymphocytes for 09/29/2005 to 03/15/2006; ratios are reported for albumin (which included prealbumin and albumin) to globulins (which included all globulins) for 09/15/2004 to 03/15/2006; albumin is reported as a concentration that was calculated by multiplying the proportion of serum protein estimated as prealbumin and albumin by biuret total protein for 09/29/2005 to 03/15/2006; and alpha 1, beta 1 and 2, and gamma globulins were each separately reported as concentrations that were calculated by multiplying the proportion of serum protein estimated as that specific globulin fraction by biuret total protein for 09/29/2005 to 03/15/2006]. Data accuracy was checked by comparing values against original data sheets. Attributes were spot checked and values were sorted to identify possible errors. For example, if data looked like a potential error, original data sheets were checked to confirm and fixed it if it was a data entry error. Additionally peer-review used sort functions in Excel to identify attributes, domains, and domain ranges. These were checked against metadata entries to ensure all definitions and levels were accounted for.
IDEXX VetTrol Controls were run on the IDEXX VetTest analyzer once per month and prior to running samples to ensure the analyzer was functioning properly. A Quality Control Panel slide was included during analyses and contained multiple chemistries for a comprehensive quality control.

Six Pacific Common Eiders were used to determine the effects of implanted transmitters. Eggs were salvaged from nests of wild birds from the Yukon–Kuskokwim Delta, Alaska, USA, and hatched at the Alaska SeaLife Center (ASLC) in Seward, Alaska, in 2003. Between hatch and October 2005, birds were housed in an outdoor aviary at the ASLC with shallow pools (<1 m deep). On October 3, 2005, birds were transferred to an outdoor seawater aquarium, in which we constructed a dive column (~ 1.5 X 1.5 m wide and 4.9 m deep) with an attached terrestrial haulout. After moving birds to the new enclosure, we trained birds to dive to the bottom of the dive column. Once birds were regularly diving to the bottom, we fed birds through a PVC pipe onto an acrylic feeding tray on the floor of the dive column. On November 30, 2005, veterinarians implanted a satellite transmitter into the abdominal cavity of each bird.

Blood-total Protein (Biuret Method): Total protein in blood serum was analyzed at the Marshfield Clinic Laboratories (Marshfield, Wisconsin, USA) using the biuret method according to the automated procedure on a Roche Modular Analytics System. The Biuret method is a colorimetric assay. The procedure is conducted in an alkaline medium, where a divalent copper reacts with protein peptide bonds forming a purple-colored biuret complex that is measured photometrically. The color intensity generated by this complex is directly proportional to the total protein concentration.

Blood-Hematology: We determined white blood cell proportions by making ≥2 blood smears directly from the collection syringe using fresh untreated blood. After drying, we stained slides using a quick dip stain system (Jorgensen Laboratories, Inc., Loveland, CO). We then manually counted cells using a microscope. Total white blood cell count was estimated using cell counts and unopette results. We determined the PCV ratio by centrifuging a capillary tube of heparinized blood for 3 minutes at 3500 rpm. We then used a Micro-Capillary Reader (IEC, Needham Heights, MA) to measure the ratio of cells to the total sample, except 1 case where the capillary tube wasn't readable and we estimated PCV by examining a spun whole blood tube. We used a refractometer to measure total protein.
