Description:
Abstract:
This data set describes nucleotide sequence data derived from 18S ribosomal DNA amplified in two fragments. A total of 87 feces from Pacific walrus and 57 marine invertebrates were examined for this study. Samples were collected from the Bering Sea and Chukchi Sea, Alaska. Samples used in the study originated from feces or muscle samples collected in the field from ice floes or benthic van Veen grab samples.
Purpose: To infer the summer diets of Pacific walrus.

Taxonomic_Procedures:
Field collected marine invertebrates were identified by skilled observers based on morphological characteristics. Operational Taxon Units (OTU) were identified genetically by comparison of DNA sequences to sequences available on NCBI Genbank using the Basic Local Alignment Search Tool (BLAST).
Taxonomic_Completeness:
Provided here is the classification of marine invertebrates to the rank of class. Further classification of individual samples and OTUs to the lowest possible taxonomic rank are provided in the table "walrus_dietMetabarcoding_taxa_alaska_sonsthagen_2012_2018.csv" included with this data package.

Attribute_Accuracy_Report:
Identifying information about samples were cross-checked with original field data. Polymerase chain reaction (PCR) amplification followed protocols described in Sonsthagen et al 2020. All laboratory techniques were performed using sterile procedures.
Logical_Consistency_Report:
Primers and adapters were trimmed using the default scoring algorithm in CLC Genomics v. 9.0 (Qiagen Inc.). Bases were trimmed on a maximum error probability of 0.01, with at most two ambiguous bases allowed and a minimum trimmed length of 50 bp. Orphaned reads were discarded. Trimmed read pairs were merged with the default merge parameters; merged reads shorter than 100 bp were discarded. See Sonsthagen et al. 2020 for additional information.

Methodology_Description:
Locus-specific primers were tailed with Illumina adaptors (P5 and P7) and indices following Nextera chemistry (Illumina, San Diego, CA). Polymerase chain reaction (PCR) amplifications were carried out in triplicate. Pooled PCR products were electrophoresed on a 1.5% agarose gel and gel purified using a MinElute Gel Extraction Kit (Qiagen) to ensure no primer dimers were present in the final library. Additional details on laboratory techniques are available in Sonsthagen et al (2020).

Entity_Type_Definition:
Table with the number of reads assigned to Operational Taxon Units (OTU) listed by walrus fecal sample. Counts are summed across OTUs with the same taxonomic rank assignment, but are not aggregated at successive ranks. That is, genus counts do not include species counts, family counts do not include genus counts etc. Blank cells indicate that an OTU could not be assigned to a particular taxonomic rank. Presented in a Comma Separated Value (CSV) formatted table.

Each attribute represents one walrus fecal sample. Attribute labels correspond to "Sample_ID" value in the table "walrus_dietMetabarcoding_sampleDetails_alaska_sonsthagen_2012_2018.csv" included with this data package. Each record provides the number of reads assigned to the OTU for each walrus fecal sample.
