Abstract:
This data set is comprised of one table with sampling information and allele sizes for 69 individual Northern Fulmars (Fulmarus glacialis) from Chagulak Island, Alaska and Hafnarey Island, Iceland. Fulmar allele sizes are provided for 10 previously published and 26 novel microsatellite loci. Additionally, allele sizes for the 26 microsatellite loci developed in this study are included for Arctic Terns (Sterna paradisaea, n=4), Common Murres (Uria aalge, n=6), Crested Auklets (Aethia cristatella, n=6), Fork-tailed Storm Petrels (Oceanodroma furcata, n=5), Marbled Murrelets (Brachyramphus marmoratus, n=5), Common Gulls (Larus canus, n=5), Pelagic cormorants (Phalacrocorax pelagicus, n=5), and Thick-billed Murres (Uria lomvia, n=5) from various locations throughout Alaska.
Purpose:
Prior cross-species screens and classical microsatellite marker development specifically in Northern Fulmar, failed to provide a multilocus marker suite adequate for use in population genetics studies.

Sampling information provided by collectors was cross-checked and verified to match the accompanying data. Sampling locations were verified with collectors, labeled, and cross-referenced to the Geographic Names Information Service (GNIS) or NGA GEOnet Names Server when possible. Quality control (QC) included a minimum of 10% re-extraction and PCR replication for each locus and all laboratory techniques were performed using sterile procedures. All duplicate characterizations of the same individual resulted in concordant data. GenBank accession numbers were cross-referenced to the appropriate individual samples as recorded. Species identification by collectors were informally verified by confirming genetic "signatures" (e.g. microsatellite size or DNA sequence) fell within expected values for that species. Field sex was not confirmed with a laboratory analysis.

For microsatellite loci, reported allele sizes were reviewed to confirm they conform to expected distributions (e.g. dinucleotide repeats with 1 base pair differences were reviewed on gel images with laboratory personnel, gaps in allele sizes were reviewed, and single occurrences of an allele size were scrutinized). Any inconsistencies in allele scores were resolved with additional replicates. Tests for null alleles and allelic dropout were implemented in MicroChecker (van Oosterhout et al 2004).

Polymerase chain reaction (PCR) amplification and scoring of microsatellite loci followed Sonsthagen et al 2004 except the thermocycler parameters consisted of a 2 min denaturation at 94 °C, followed 40 cycles at 94°C for 15 seconds, 50°C for 15 seconds and 72 °C for 30 seconds with a final extension at 72 °C for 30 min. Microsatellite alleles were resolved on a 25 cm 6% polyacrylamide gel using a LI-COR (IR 4200) automated DNA sequencer. For quality control purposes, we extracted, amplified, and sequenced, 10% of the samples in duplicate.

This buffer is often used to preserve non-blood tissue samples collected for DNA analysis at ambient temperatures until DNA extraction can occur. Current and past personnel of the Alaska Science Center as well as many associates familiar with the buffer are unsure of its origins, though its use at the Alaska Science Center for tissue preservation is known from at least 1996. Its formulation is (4.00 M Urea, 0.2 M NaCl, 10 mM EDTA, 0.5% N-Lauroyl-Sarcosine, 100 mM Tris-HCl, pH 8.0) stored at room temperature.

Initial microsatellite allele sizes were determined by scoring based on PCR product size relative to an M13 DNA sequence ladder using Gene Profiler 4.05 (Scanalytics, Inc., Fairfax, VA). Subsequent gels included these known size individuals from which relative sizes of unknown individuals were assigned using Gene Profiler 4.05. For this reason, these microsatellite data cannot be combined with data generated outside the USGS Alaska Science Center without molecular size calibration between laboratories. Tests for null alleles and allelic dropout were implemented in MicroChecker (van Oosterhout et al 2004).
